<p><span style="color: #800000;"><strong>C-H活性化によるアリールシクロプロピルアミン誘導体の合成</strong></span></p>
<p>アリールシクロプロピルアミンは、医薬品など生物活性化合物の部分構造として利用される骨格である。一般的には、芳香族アルデヒドのWittig反応によって合成した桂皮酸誘導体のシクロプロパン化、Curtius 転位を経て合成される。しかしながら、この従来法を用いて、多様なアリール基を有するシクロプロピルアミンを合成するためには、出発物質の桂皮酸誘導体の合成を含めて多段階反応が必要となるため、より短工程での効率的な合成ルートが望まれる。</p>
<p>今回我々は、市販のシクロプロピルアミンを出発原料として、イリジウム触媒を用いたシクロプロパン環の sp<sup>3</sup>C–H 結合直接ボリル化、続くパラジウム触媒を用いたヨウ化アリールとの鈴木−宮浦クロスカップリング反応を経て、短工程で、多種多様なアリールシクロプロピルアミンを合成することに成功した。シクロプロピルアミンのC–H ボリル化はシス選択的に進行する。一方で、鈴木−宮浦クロスカップリングは、窒素下でトランス体、酸素化でシス体優位に生成することも見出した。単純に窒素、酸素を変えるだけで、トランス及びシスのアリールシクロプロピルアミンにアクセス可能である。</p>
